Esomeprazole is a proton pump inhibitor (PPI) that specifically inhibits the proton pump of the gastric parietal cell, the final stage of gastric acid secretion. It is administered as a prodrug, activated by sulfitase, an enzyme present in the gut, which transforms esomeprazole into its active form. This activation increases the drug’s lipophilicity, allowing it to cross the cell membrane and bind to the proton pump. Esomeprazole is a potent and efficient inhibitor of the gastric proton pump, H+/K+ ATPase, which leads to a decrease in gastric acidity and increased pH levels. Esomeprazole is indicated for the treatment of symptoms associated with gastroesophageal reflux disease (GERD), the healing of erosive esophagitis, treatment of gastric ulcers, in combination with other treatments, Helicobacter pylori infection, and the management of Zollinger-Ellison syndrome.
